412元件插入引起的棒眼果蝇杂交后代scarlet基因突变分析
  • 摘要

    设计了6对引物对棒眼果蝇(Drosophila melanogaster)Bar与野生型果蝇杂交所产生的亮红眼突变型果蝇stbr的scarlet基因进行了测序,结果在scarlet基因的第4外显子检测到了1个7.5kb的插入,进一步的测序证实此7.5kb插入为逆转座子412元件.在对棒眼果蝇和从日本引进的眼色突变品系st1果蝇的scarlet基因测序分析时也都检测到了412元件的插入,不同的是棒眼果蝇scarlet基因中412插入呈杂合型.对所有的412插入位点分析时发现,stbr果蝇的412元件以与scarlet基因转录一致的方向插入到了其第4外显子的1612th~1613th bp之间,且412元件的5'长末端重复LTR区发生了450 bp的缺失;stl果蝇的412元件则以与scarlet基因转录相反的方向插入到了其第4外显子的1722nd~1723nd p之间.
